异养小球藻的筛选鉴定及其特性研究

Study on Isolation and Identification of a Strain Heterotrophic chlorella and its Growth Characteristics

  • 摘要: 在天然水体中筛选得到1株能高速异养生长的藻株WLZ-H。用扫描电镜对藻株进行细胞形态分析,并结合16S rDNA分子生物学鉴定,初步确定藻株WLZ-H属于小球藻属(Chlorella sorokiniana)。对藻株WLZ-H进行环境因子单因素试验,发现KNO3和葡萄糖的浓度对藻株WLZ-H生长影响显著;过高的温度会降低藻株WLZ-H的酶活性,从而降低其生长速率;pH会随着藻株WLZ-H的生长而变化,并达到适合藻株WLZ-H每个时期生长的pH。藻株WLZ-H的最佳生长条件:初始pH为7.0,KNO3浓度为1.5 g/L,葡萄糖浓度为40 g/L,温度31 ℃。

     

    Abstract: A heterotrophic WLZ-H strain growing fast was screened from natural waters. The cell morphology of the alga strain was analyzed with scanning electron microscope and, in combination with 16S rDNA molecular biology identification method, it was preliminarily conclude that the WLZ-H strain belonged to Chlorella sorokiniana.Single environmental factor test on the WLZ-H strain showed that the concentration of KNO3 and glucose had significant effects on WLZ-H strain growth; too high temperature would decrease enzyme activity of WLZ-H strain and consequently decrease its growth; pH would change with the growth of WLZ-H strain and reach the suitable value in different growth phase.The optimal growth conditions of WLZ-H strain were:initial pH 7.0, KNO3 content 1.5 g/L, glucose content 40 g/L, temperature 31 ℃.
